Abstract

Background

To better understand the molecular basis of herpes zoster (HSV) and the role of antiviral agents in the treatment of HSV-associated genital herpes (USGHS). We assessed the pharmacological and toxicological effects of two novel oral antivirals (5-amoxicillin-glutamic acid and aciclovir) in a randomised, double-blind, placebo-controlled study of a novel oral regimen of oral aciclovir in adults with HSV-2 or HSV-1 infection. Methods: This study was an in vitro study to assess antiviral activity and the pharmacokinetics of the novel oral aciclovir in healthy volunteers. Results: There was a significant increase in the AUC and AUC0-1 levels of 5-amoxicillin-glutamic acid from AUC0-1 to AUC0-1 and a significant increase in the AUC and AUC0-1 concentrations of aciclovir from AUC0-1 to AUC0-1, in the group of participants who received treatment with 5-amoxicillin-glutamic acid, compared to the group who received treatment with placebo. Conclusions: This study showed that both the novel aciclovir and the active metabolites of aciclovir were bioequivalent to 5-amoxicillin-glutamic acid and aciclovir in healthy volunteers. This study also revealed that oral aciclovir was effective in reducing the risk of recurrent genital herpes in adults with HSV infection. Future research should focus on reducing the risk of recurrent genital herpes in patients with HSV infection, to identify effective treatment and to develop a regimen with safe and effective oral aciclovir in patients with HSV infection.
